The effect of 5-fluorouracil on DNA-thymine synthesis in human tumors.

چکیده
eluent. Uptake patterns were highly individual, and large differences were noted both in the baseline uptakes and in the effect of 5-fluorouracil on precursor incorporation. Neither tissue of origin nor control uptake influenced the in vitro response to the drug. A decrease in formate incorporation was usually associated with a reciprocal increase in thymidine incorporation. Clinical data concerning tumor response were available for seven of the patients studied. The clinical response appeared to correlate with the in vitro behavior of these tumors.

Response of DNA thymine synthesis in human tumor and normal tissue to 5-fluorouracil.
Thymidine-methyl-3 H and either formate-' @ C or @ C were added to incubations containing various concentrations of 5-fluorouracil. The thymine in DNA was isolated, and precursor incorporation was determined by dual-label liquid scintillation counting. MATERIALS AND METHODS The methods used in this study have been previously described (12). In summary, tissues obtained from patients who had not...
متن کاملPyrimidine studies. III. Effect of several compounds with antitumor activity on utilization of precursors for synthesis of nucleic acid pyrimidines.
Addition of amethopterin to an incubation medium containing slices of the human tumor transplants H.S. #1 and II.Ep. #3 decreased selectively the utilization of orotie acid-C 1~ for the synthesis of DNA thymine. A substantially larger depression of the specific activity of DNA thymine was observed upon addition of both amethopterin and 5-bromouridine to the incubation medium than upon addition ...
متن کاملEffect of 5-bromodeoxyuridine on deoxyribonucleic acid-thymine synthesis and cell metabolism of lymphatic tissues and tumors.
5-Bromodeoxyuridine (BrUDr) is an effective inhibitor of the growth of E. coli and Lactobacitti, and this growth inhibition can be reversed by thymidine (1-3). There is evidence that this thymidine analog can replace the thymine of both E. coli and bacteriophage deoxyribonucleic acid (DNA) on an equimolar basis (7, 11, 12). In E. coli, nearly 50 per cent of the thymine can be replaced. Whereas ...
متن کاملRole of catabolism in pyrimidine utilization for nucleic acid synthesis in vivo.
The incorporation of pyrimidines into nucleic acids in vivo is increased by inhibition of pyrimidine catabolism with diazouracil. The utilization of iodouracil or thymine for DNA synthesis can be increased approximately 20-fold by simultaneous administration of diazouracil and a purine deoxyribonucleoside. The incorporation of iodouracil and thymine, when administered at high doses, is elevated...
متن کاملInhibitory effect of halogenated pyrimidines on multiplication of viruses in tissue culture systems.
It is of interest to examine the inhibitory effect of halogenated pyrimidines on the multiplication of viruses in tissue culture systems, in view of finding the fundamental knowledge to obtain antiviral drugs. Concerning the inhibition of biosynthesis of nucleic acid by halogenated pyrimidines, Heiderberg and his coworkers(') found that 5-fluorouracil inhibited the process of biosynthesis of RN...
